Complete Medium:
For arginine broth, lysine (Falkow) broth or ornithine broth, add 5.0 g of L-arginine, L-lysine or L-ornithine to 1.0 L of base. Adjust the pH so that the value after sterilization is 6.5 " 0.2. Dispense 5.0 mL into 16x125 mm screw-cap tubes. Autoclave the loosely capped tubes for 10 min at 121oC. Screw caps on tightly for storing and after inoculation. For control use unsupplemented base.
*When using for halophilic Vibrio spp., add NaCl to a final concentration of 2-3%.

Weigh ingredients into 1.0 L of distilled water. Dispense into bottles and sterilize at 121oC for 15 min.

Complete Medium:
Add components, except chloramphenicol solution, to distilled water and bring the volume to 990 mL. Mix thoroughly. Gently heat and bring to a boil. Autoclave at 121oC for 15 min. Aseptically add filter sterlized chloramphenicol solution and mix thoroughly. Pour into plates.

Complete Medium:
Add components, except glycerol, to 800 mL distilled water. Steam to dissolve before adding water to 1.0 L. Add glycerol and autoclave at 121oC for 15 min.

Complete Medium:
Dilute the stock buffer solution 20 fold, (e.g., 5.0 mL buffer solution made up to 100 mL with distilled water). If the diluted stock solution is going to be held longer than one working day prior to use, it should be sterilized at 121oC for 15 min in suitable bottles.
Prepare the chlorine solution by adding 10.0 mL of a commercial chlorine solution (e.g. Javex™ at 5.25% available chlorine or other suitable chlorine solution of comparable strength) to each litre of the diluted buffer (1/20M) solution. The final available chlorine concentration should be between 500 and 525 ppm and the final pH 6.5. The available chlorine should not be less than 100 ppm.
