Glossary of agars, broths and other reagents

Neutralizing Buffer (Environmental sampling)
Basal Medium
Potassium phosphate monobasic 0.0425 g
Sodium thiosulphate 0.16 g
Aryl sulfonate complex 5.0 g
Sodium hydroxide (NaOH) 0.008 g
Distilled water 1.0 L
pH 7.2 ± 0.2  

Complete Medium:

Weigh ingredients into 1.0 L of distilled water.Mix and dispense into bottles. Sterilize at 121oC for 15 min.

Nitrate Motility (Nm) Agar(Motility-Nitrate Medium, Buffered for C. perfringens)
Basal Medium
Peptone 5.0 g
Beef extract 3.0 g
Potassium nitrate 1.0 g
Disodium phosphate 2.5 g
Galactose 5.0 g
Glycerol 5.0 g
Agar 3.0 g
Distilled water 1.0 L
pH 7.3 ± 0.2  

Complete Medium:

Add components, except agar, to distilled water. Mix thoroughly and adjust pH. Add agar and gently heat to boiling to dissolve. Dispense into tubes and autoclave at 121oC for 15 min. If not used within 4 h, deaerate by heating for 10 min in boiling water or flowing steam and cool.

Nitrate Reagent
Basal Medium
5-amino-2-naphthalene sulfonic acid (5-2 ANSA) solution :  
5-2 ANSA 0.1 g
Acetic acid (15% aqueous solution of glacial acetic acid) 100 mL
Sulfanilic acid solution :  
Sulfanilic acid 0.4 g
Acetic acid (15% aqueous solution of glacial acetic acid) 100 mL

Complete Medium:

Dissolve the 5-2 ANSA and sulfanilic acid in 15% acetic acid. Filter both solutions through filter paper (Whatman # 41 or equivalent), and store the reagents separately in tightly stoppered brown bottles at 4oC. Mix equal amounts as needed. Discard unused reagent after 6 months.

Nutrient Agar (plates or slants)
Basal Medium
Peptone 5.0 g
Beef extract 3.0 g
Agar 15.0 g
Distilled water 1.0 L
pH 6.8 ± 0.2  

Complete Medium:

Suspend ingredients in 1.0 L of distilled water and heat to boiling with frequent swirling. Dispense into tubes or plates and autoclave at 121oC for 15 min. Cool tubes in a slanted position. Plates can be stored for up to 8 weeks at 4oC.

Nutrient Broth (NB)
Basal Medium
Peptone 5.0 g
Beef extract 3.0 g
Distilled water 1.0 L
pH 6.8 ± 0.2  

Complete Medium:

Dissolve ingredients in 1.0 L of distilled water. Dispense into appropriate glassware and autoclave at 121oC for 15 min.
