Acid Broth(AB)
Basal Medium
Proteose peptone 5.0 g
Yeast Extract 5.0 g
Dextrose 5.0 g
Potassium phosphate, dibasic (K 2HPO 4) 4.0 g
Distilled water 1. 0 L

Complete Medium

Adjust the pH of the medium to the pH of the food being analysed with 1N HCl solution. Sterilize at 121oC for 20 min.

Aeromonas Medium (Ryan's)
Basal Medium
Sodium Thiosulfate (Na 2S 2O 3) 10.67 g
Proteose peptone 5.0 g
Sodium chloride (NaCl) 5.0 g
Xylose 3.75 g
L-Lysine.HCl 3.5 g
Yeast extract 3.0 g
Sorbitol 3.0 g
Bile salts No.3 3.0 g
Inositol 2.5 g
L-Arginine.HCl 2.0 g
Lactose 1.5 g
Ferric ammonium citrate 0.8 g
Bromthymol blue 0.04 g
Thymol blue 0.04 g

Supplement
Ampicillin 5. 0 mg

Complete Medium:

Add components, except ampicillin, to distilled water. Mix thoroughly. Gently heat and bring to a boil. Do not autoclave. Cool to 50oC. Aseptically add 5.0 mg of ampicillin and dispense.

Note: The supplement is commercially-available.

AKI Medium
Basal Medium
Peptone 15.0 g
Yeast extract 4.0 g
Sodium chloride (NaCl) 5.0 g
Distilled water 970 mL
pH 7.4 ± 0.2  
Supplement
Sodium Bicarbonate (NaHCO 3), 10% aqueous, filter sterilized 30.0 mL

Complete Medium

Prepare on day of use. Dissolve first 3 ingredients in distilled water. Autoclave 15 min at 121oC . Cool then add 30 mL NaHCO 3. Mix and aseptically dispense into screw-cap tubes (15.0 mL for 16x125 mm tubes).

Alkaline Peptone Salt Broth (APS)
Basal Medium
Peptone 10.0 g
Sodium chloride (NaCl) 30. 0 g
Distilled water 1.0 L
pH 8.5 ± 0.2  

Complete Medium:

Add components to distilled water. Mix thoroughly. Adjust the pH so that the value after sterilization is 8.5 ± 0.2. Dispense 10 mL into tubes and autoclave 10 min at 121oC.

Alkaline Peptone Water (APW)
Basal Medium
Peptone 10.0 g
Sodium chloride (NaCl) 10.0 g
Distilled water 1.0 L
pH 8.5 ± 0.2  

Complete Medium:

Add components to distilled water. Mix thoroughly. Adjust the pH so that the value after sterilization is 8.5 ± 0.2. Dispense and autoclave 10 min at 121oC.

Arginine Glucose Slant (AGS)
Basal Medium
Peptone 5.0 g
Yeast extract 3.0 g
Tryptone 10.0 g
Sodium chloride ( NaCl ) 20.0 g
Glucose 1. 0 g
L-Arginine (hydrochloride) 5.0 g
Ferric ammonium citrate 0.5 g
Sodium thiosulfate 0.3 g
Bromcresol purple 0.02 g
Agar 13.5 g
Distilled water 1.0 L
pH 6.9 ± 0.1  

Complete Medium:

Add ingredients to distilled water. Mix thoroughly and heat to dissolve. Dispense into tubes and autoclave at 121o C for 10-12 min. Allow tubes to cool in a slanted position.
